Acridine Orange Staining

Figures (A), (B) and (C) were the sperm samples stained with Acridine Orange Dye to identify normal (double-stranded DNA) or abnormal (single-stranded DNA) sperm chromatin. This technique was processed under fluorescence microscopy.

  • Figure (A): This sperm sample was collected from a fertile gentleman. There was a 38% sperm head appeared in yellow to red color, i.e., single-stranded and abnormal DNA. The other sperm showed green head, i.e., double-stranded and normal DNA. (Note: Only green-colored sperm head is normal.)

  • Figure (B): This sperm sample was collected from an infertile patient. You can see that almost all of the sperm heads appeared in orange color, i.e., single-stranded and abnormal DNA.

  • Figure (C): Almost all of sperm cells (about 99%) appeared in green color, i.e., double-stranded and normal DNA, following sperm micro-separation via Wang Tube Technology.
